Private labeling adds a packaging and subdivision process after the peptide has already passed bulk testing. That step can introduce mix-ups, concentration errors, moisture exposure and uneven fills. A robust private label peptide vial filling quality control plan links the released bulk lot to every finished vial through reconciliation, in-process checks, label control and representative finished-unit testing.
Approve the master record before components arrive
The record should identify peptide, sequence or material code, salt form, target net content, excipients, solution concentration if filled before lyophilization, vial and stopper specification, fill volume or mass, lyophilization cycle, label text, pack configuration and sampling plan. Marketing artwork is not a manufacturing instruction.
Clarify whether the label claim is net peptide, gross powder or solution-equivalent content. The bulk assay, water and counterion values determine how much powder or stock solution is required. Using gross bulk mass as if it were pure peptide creates a systematic underfill even when the equipment performs perfectly.
Line clearance prevents the most damaging errors
Before filling, remove prior product, labels, cartons, caps and documents from the room and equipment. Record the inspection. Similar blue-capped vials and near-identical peptide names create high mix-up risk. Barcode or electronic verification helps only if the master data are correct and operators cannot bypass it.
Segregate printed components by product and lot. Reconcile issued, used, damaged and returned labels. Unexplained excess or shortage should stop release. A correct vial with the wrong strength or batch label is not a cosmetic defect; it breaks traceability.
Bulk reconciliation detects hidden loss
Record starting quantity, samples, transfer loss, equipment hold-up, rejects, filled output and remaining material. Calculate theoretical and actual yield using the same content basis. Large unexplained loss can signal spills, adsorption, wrong concentration, poor transfer or undocumented diversion.
For solution filling, verify concentration after final volume adjustment and mixing. Sample from more than one location when tank uniformity is not established. A clear solution can still have a concentration gradient if mixing and addition order are poorly controlled.
Fill weight is not peptide content
Checkweighing is useful for monitoring mechanical consistency, but a vial’s total weight includes container variation and all formulation components. Establish tare strategy and balance capability. For low fill masses, gravimetric noise may be large relative to the peptide quantity.
Finished-unit assay or a validated solution-fill calculation may be required to confirm peptide amount. Sample the beginning, middle and end of the run, plus random units. Average compliance cannot excuse an individual distribution problem when the specification requires uniform units.
Lyophilization creates additional variability
Loading pattern, shelf temperature, chamber pressure, primary drying and secondary drying influence residual moisture and cake structure. Edge and center vials may experience different heat transfer. Map the dryer and include locations with expected extremes in qualification and routine sampling.
Cake appearance should be recorded, but it is not a substitute for moisture, assay or purity. Collapsed cakes can sometimes meet chemical specifications; attractive cakes can still have excessive moisture. Define defect categories and analytical disposition rules before inspection begins.
Container closure and post-fill handling
Control stopper placement, crimp dimensions, cosmetic defects and container-closure integrity where required. Vials that lose vacuum or admit moisture may degrade during shipping even when release testing was performed promptly. Secondary packaging should protect against breakage, light and label damage.
Limit room-temperature exposure of bulk and open vials. Record cumulative time, not only the longest single interval. Repeated short holds during filling, inspection and labeling can exceed the qualified exposure.

Should the finished vials carry the same COA as the bulk powder? The bulk data can be referenced, but tests affected by filling, lyophilization or packaging need representative finished-unit evidence. The certificate should make the sampling stage clear.
Before approving a private-label partner, review one executed batch record with confidential values redacted, inspect how labels are reconciled and commission independent tests on randomly selected vials. The most dependable operation can explain every gram of bulk and every numbered unit from receipt through final carton.
