Peptide KLH and BSA Conjugation Service Buyer Guide

A peptide-carrier conjugate is inherently more heterogeneous than a defined synthetic peptide. The practical release questions are attachment site, loading distribution, removal of free peptide and preservation of the required carrier controls. A peptide KLH BSA conjugation service should describe the chemistry and calculation basis rather than report only milligrams of recovered conjugate.

Design the peptide with one intended handle

An added terminal cysteine enables thiol-selective coupling when the native sequence lacks accessible thiols. Amine-based coupling can involve the N-terminus and lysine side chains, producing less defined orientation. State the linker, handle position and whether the added residue belongs to the assay epitope.

Protect or account for native cysteines and lysines that could create multiple attachment sites. Order unconjugated peptide from the same lot for analytical controls.

Choose carrier by downstream use

KLH offers high molecular size and many attachment sites but is structurally heterogeneous. BSA and OVA can be useful as assay or alternate carriers. Avoid using the same carrier for both immunization and detection when carrier-directed responses could interfere with interpretation.

Loading ratio is an estimate with a method

Peptide-to-carrier ratio may be inferred from starting materials, UV measurements, amino-acid analysis or other assays. Each approach has assumptions. A reaction input ratio is not a measured final loading ratio. Ask for the calculation, recovery and uncertainty.

Large carriers produce broad SEC or electrophoretic patterns. Mass spectrometry may not yield a simple intact conjugate mass. The analytical package should fit the carrier rather than imitate a small-peptide COA.

Remove free peptide and reagent

Desalting, dialysis or SEC can remove small molecules, but recovery and cutoff must be demonstrated. Residual free peptide can create assay signal unrelated to the conjugate. Residual crosslinker or reducing reagent can affect storage and downstream testing.

Preserve traceability between components

The batch record should link peptide lot, carrier lot, conjugation reagent, reaction conditions, purification and final aliquots. The label needs the carrier identity and conjugate lot, not only the peptide name. Store unconjugated peptide and carrier control separately.

Define delivery by usable material

Specify buffer, concentration, carrier content basis, preservative, aliquot size, storage and shipping. Avoid repeated freeze-thaw. If sterile filtration is requested, check filter recovery because large conjugates can adsorb.

  • Measured or estimated loading method.
  • Free peptide removal and final buffer.

Can HPLC purity be reported for a KLH conjugate? A conventional peptide HPLC percentage is generally not an adequate description of the heterogeneous carrier product. Request component identity, loading evidence, purification records and suitable protein-level characterization. Send the intended carrier and assay format with the RFQ so the supplier can design both conjugate and controls coherently.
