A frozen label does not guarantee a controlled shipment. For lyophilized peptide cold-chain shipping stability, the useful question is not whether the parcel felt cold on arrival; it is whether the product experienced a time-temperature-humidity profile that the packaging study and molecule-specific data can support. Freeze-dried peptides are often more stable than solutions, yet oxidation, deamidation, aggregation, moisture uptake, and container leakage do not stop simply because a vial contains a dry cake.
Separate product temperature from box temperature
Courier tracking records ambient events. A data logger positioned against dry ice records a local cold spot. Neither necessarily represents the vial. During shipping qualification, place probes near the warmest predicted product position, normally close to an outer wall or lid seam, and in the central payload. The difference between these locations matters during customs holds, weekend storage, and repeated opening at transfer hubs.
Ask the supplier for the validated shipping duration, payload configuration, coolant quantity, season, and acceptance range. “Ships with ice packs” is not a protocol. Gel packs designed for 2–8°C behave differently from dry ice near −78°C. Some lyophilized materials tolerate short controlled ambient transit, while others require protection from heat or moisture more than continuous subzero temperature. The shipping condition should follow evidence, not a catalogue-wide habit.
What to check before dispatch
The vial-closure system is the first barrier. Inspect crimp integrity, stopper seating, glass compatibility, headspace control, and evidence of vacuum where specified. A weak seal allows moisture ingress as the parcel warms and cools. Secondary packaging should protect against condensation and prevent direct contact between vials and coolant. Direct dry-ice contact can create extreme local temperatures and brittle packaging, while thawing gel packs can soak labels and compromise traceability.
Request a packing map and logger serial number before collection. Confirm who activates the logger, where it is placed, the logging interval, time zone, and alarm limits. A 30-minute interval may miss short transfers in a hot loading area; a one-minute interval creates more data but does not repair poor placement. Five to ten minutes is often practical for international routes, provided the memory capacity exceeds the expected duration.
Customs delays are part of the route
International peptide logistics fail at borders more often than in aircraft holds. A route qualified for 72 hours is unsuitable when typical clearance takes four days. Add a contingency period based on lane history, not the courier’s best-case estimate. Confirm whether replacement coolant can be added during a hold and whether the forwarder is authorized to handle dry ice. Dry ice is regulated as dangerous goods for air transport; documentation and quantity marking must be correct.
Hidden cold-chain charges also deserve attention. Quotes may exclude dry-ice replenishment, dangerous-goods handling, remote-area fees, logger recovery, customs storage, brokerage, or return transport. Ask for a landed logistics quotation with each conditional fee identified. A cheap freight line can become the most expensive option after a three-day inspection hold.
Receiving procedure: do not open first and investigate later
Record the arrival time, external condition, seal status, remaining coolant, and logger state before moving the package. Photograph all sides and the internal packing sequence as it is opened. Download the logger immediately and preserve the native file, PDF report, serial number, and checksum if available. Do not discard packaging until disposition is complete.
Inspect vials under consistent lighting. Note cracked glass, displaced stoppers, loosened crimps, collapsed or melted cake, splashing marks, unusual color, and particles. Cake appearance alone cannot establish chemical stability. A neat white plug may still contain oxidized or deamidated peptide, while a cosmetically irregular cake may meet analytical specifications. Visual inspection is a triage tool, not a release assay.
How should a temperature excursion be assessed?
Do not reject or accept solely from the highest recorded temperature. Evaluate the duration, product position, known stability profile, packaging configuration, and cumulative exposure. A brief 12°C reading at the outer probe is different from 24 hours at 30°C across both probes. Compare the event with excursion or accelerated-stability data for the same formulation, container, salt form, and moisture range.
If evidence is insufficient, quarantine the lot and test representative units. HPLC can detect increased related substances, and LC-MS can help identify oxidation or cleavage products. Water determination is valuable after suspected seal or humidity exposure. Functional or binding assays may be appropriate for complex peptides when chemical purity alone does not demonstrate retained performance. Do not “average out” an alarm by combining cold and warm periods.
Why lyophilized does not mean indestructible
Residual moisture strongly affects mobility and degradation in the solid state. Oxygen in the headspace can matter for methionine-, tryptophan-, or cysteine-containing sequences. Repeated temperature cycling may change cake structure or encourage stopper movement. Lipidated peptides present an additional problem: aggregation and surface interactions can depend on excipients and reconstitution history, and standard short-term HPLC data may not predict long-route behavior.
Once reconstituted, the risk profile changes sharply. Solutions generally offer greater molecular mobility and more opportunity for hydrolysis, oxidation, adsorption, and microbial contamination. - Lane or packaging qualification with summer and winter challenge conditions.
- Payload map, coolant loading, logger placement and validated duration.
- Real-time and accelerated stability results with analytical methods.

- Container-closure and residual-moisture specifications.
- Itemized freight, dangerous-goods, customs and replenishment fees.
For a first international order, run a pilot shipment using the exact commercial pack-out and at least two loggers. Agree on arrival testing and excursion decisions before dispatch. A supplier willing to share lane assumptions, rather than merely promise “cold chain,” gives the buyer a defensible basis for accepting or rejecting the lot.
